Give 2 strong points (characteristics) about yourself & 2 weak points. Focus on work related traits.
The weak points are usually harder to come up with during the interview, and they tell the interviewer more about you, how to supervise you, how to utilize you and how humble and honest you might be.
I like to see that a person has actually done some real useful homework on the position/company and city/area that they are applying for. Don't just read their webpage!

at the end of the interview as we were shaking hands..."btw, do you know the CEO of this companies name?"
I did, but I wouldn't have expected to be asked that...I have also been asked "what's our stock looking like now"
Anything that focuses on weak points, like Jilly said. When I interview, I always ask a question like that. It is usually a home run for the potential employee, or a giant bomb.
I also like "have you ever stolen anything from a previous employer?" Everyone has walked out with company equiptment, only the truely honest ones remember it. Pens, Notepads, little stuff that no one cares about.
Had one guy tell me he took a computer once....wonder if I hired him?
Go to a bookstore and get a book on interview questions, there will be some real tough ones in there.
Make sure to dress above the position, look the interviewer in the eye (don't stare), and don't talk to much...give them a chance to take notes, you don't need to fill every second with your voice. Be consise, clear, on-point, and ASK for the position at some point. "Sir/Maam, I know I am a great fit for your company, I want you to know that I want this position, how can I have this position" Or just ask. "Can I have this position""I want to know God's thoughts, the rest are just details"

Toughest question I ever had was: Why should we hire you instead of one of the other 250 people applying for this job? I guess it goes back to the previous post about explaining your strengths vs weaknesses.
Guess it worked....been at my company 23 years.
Either that or the hiring board was drunk
MikeAmbivalent? Yes. Or Not.
